ABSTRACT

<p><b>OBJECTIVE</b>To characterize the specific monoclonal antibodies to Aspergillus conidia.</p><p><b>METHODS</b>Flow cytometry was used to examine the reactivity of the specific monoclonal antibodies to Aspergillus conidia.</p><p><b>RESULTS</b>Both the monoclonal antibodies MA3 and Con2 showed specific reactivity to Aspergillus conidia suspensions. MA3 was capable of binding to the conidia of A.fumigatus, A.flavus, A.niger and A.terreus, while Con2 was reactive only to the conidia of A.fumigatus.</p><p><b>CONCLUSION</b>Two specific monoclonal antibodies (MA3 and Con2) to Aspergillus conidia have been obtained.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To establish a method for inducing apoptosis of rhesus peripheral blood lymphocytes (PBLs).</p><p><b>METHODS</b>Rhesus PBLs were irradiated with X-ray, (60)Co gamma-rays and ultraviolet (UVC254 nm), respectively, and the cell apoptosis was evaluated with flow cytometry using annexin-V staining and propidium iodide staining.</p><p><b>RESULTS</b>X-ray and (60)Co gamma-ray irradiation induced only low apoptotic rates of the PBLs, and UVC resulted in the highest apoptotic rate of about 60%. UVC irradiation of the PBLs in RPMI supplemented with 10% heat-inactivated fetal calf serum for 60 min at a distance of 20 cm led to an early apoptotic rate of 58.85% and necrotic rate of 11.5%. The apoptotic rate of PBLs increased in a dose- and time-dependent fashion.</p><p><b>CONCLUSION</b>For inducing apoptosis of the rhesus PBLs, UVC can be more effective than X-ray and (60)Co gamma-ray. The highest apoptotic rate can be achieved when the rhesus PBLs in RPMI supplemented with 10% heat-inactivated fetal calf serum are exposed to UVC for 60 min at the distance of 20 cm.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To establish an effective method for purification of Helicobacter pylori UreB fragment and conduct functional analysis of the purified protein.</p><p><b>METHODS</b>The protein fragment expression was induced by IPTG and the expressed protein was purified through affinity chromatography and ion-exchange chromatography. The purity of the fragment was determined by high-performance liquid chromatography (HPLC), and the specific biological activity of the purified fragment was assayed by urease activity inhibition test.</p><p><b>RESULTS</b>The protein fragment was highly expressed in E. coli with a purity over 91%. The protein fragment showed highly specific biological activity and the specific antibody induced by this fragment in rabbits could inhibit the activity of urease in a dose-dependent manner.</p><p><b>CONCLUSION</b>The UreB fragment with high purity and biological activity can be applied for further studies.</p>

ABSTRACT

Objective To explore the techniques of renal transplantation in diabetic recipients with lilac atherosclerosis.Methods Overall,54 renal transplantations were performed in 51 diabetic recipients with lilac atherosclerosis.The renal arteries were end-side anastomosed to iliac arteries with atherectomy in 19 cases and without atherectomy in 13 cases,or end-end anastomosed to internal iliac arteries through titani- um stapler after atherectomy in 22 cases.Results Primary nonfunetion of renal allografts occurred in 3 cases due to poor blood perfusion of the renal grafts;delayed graft function occurred in 9 cases(17.6%), and the other 42 grafts functioned well immediately after transplantation.Two patients died of sudden cardiac arrest during the perioperative period.During a follow-up of 11-70 months,the 1-year and 3-year survival rates of recipients/grafts were 89.8%/87.8% and 84.4%/81.3% ,respectively.The main cause of graft loss was recipients' death with functioning graft.Conclusions The artery anastomosis of renal graft in di- abetic recipients with lilac atherosclerosis is difficult to perform.In order to achieve enough blood peffusion of the renal graft,it is important to select proper artery for anastomosis and to perform atherectomy.The recipi- ents with coronary artery disease should receive myocardial revascularization routinely before transplantation.
